GNRHR

GNRHR, also known as Gonadotropin-Releasing Hormone Receptor, and by other names such as GRHR, GnRH receptor, or GnRH-R, is a G protein-coupled receptor (GPCR) primarily expressed in the anterior pituitary gland. It plays a key role in regulating the release of reproductive hormones. The mechanism of action for GNRHR involves binding to gonadotropin-releasing hormone (GnRH), which activates the receptor. This activation leads to G protein-mediated signaling, increasing intracellular cyclic AMP (cAMP) levels and promoting the release of luteinizing hormone (LH) and follicle-stimulating hormone (FSH). GNRHR not only plays a critical role in the reproductive system but is also associated with the development of various diseases. For example, in breast cancer, the expression of GNRHR correlates with clinical and pathological characteristics and prognosis. In particular, high expression of GNRHR may be associated with improved disease-free survival (DFS) in hormone receptor-negative breast cancer.

Additionally, GNRHR signaling in pituitary cells involves the activation of the mitogen-activated protein kinase (MAPK) cascade, providing an important link for signal transduction from the cell surface to the nucleus and playing a role in the transcriptional regulation of gonadotropins. GNRHR is also expressed in non-pituitary cells and has been found to inhibit cell proliferation in some cell types. Therefore, GNRHR represents a potential target for GnRH analogs used in cancer therapy.
