Description
TNFRSF1B (p75/TNF-RII) serves as a critical decoy receptor modulating TNF-α signaling in inflammatory and tumor microenvironments, making it an essential tool for competitive inhibition assays and therapeutic antibody screening. This tag-free recombinant construct spans the extracellular ligand-binding domain (residues 24–206) and demonstrates robust neutralizing activity, with an ED50 below 0.2 μg/mL in TNF-α-mediated L-929 cytotoxicity assays, corresponding to a specific activity exceeding 5,000 IU/mg. The confirmed ligand-blocking function supports use in receptor-ligand interaction studies by ELISA or SPR, blocking antibody screening, epitope mapping for biologic drug candidates, and as a positive control in TNF-α binding assays. Expressed in *E. coli* without affinity tags, this protein provides a suitable basis for small-molecule inhibitor screening, with purity exceeding 97% by SDS-PAGE and endotoxin levels below 1.0 EU/μg satisfying criteria typical for cell-based functional assays.
