Description
TNF-α drives NF-κB and MAPK signaling cascades central to inflammation, apoptosis, and tumor microenvironment remodeling, making cytotoxic potency and endotoxin control essential for meaningful in vitro results. This recombinant human TNF (residues 77–233), expressed in *E. coli* with an N-terminal 6×His tag, delivers an ED50 below 0.05 ng/ml in the murine L929 cytotoxicity assay—corresponding to a specific activity exceeding 2.0 × 10⁷ IU/mg—providing the signaling potency required for NF-κB reporter assays, caspase-dependent apoptosis studies, and dose–response standardization in cytokine detection platforms. Endotoxin levels below 1.0 EU/μg eliminate LPS-driven artifacts that confound macrophage activation and primary immune cell experiments, while purity above 97% by SDS-PAGE supports use as a reliable reference standard, ELISA coating antigen, or functional ligand in tumor biology animal models.
