Description
TNF-α drives NF-κB and MAPK signaling cascades central to inflammation, apoptosis, and tumor microenvironment remodeling, making precise functional activity essential for meaningful in vitro results. This recombinant human TNF-α (residues 77–233) demonstrates an ED50 of 10–40 pg/mL in L-929 cytotoxicity assays with actinomycin D, confirming high signaling potency suitable for apoptosis and cell viability studies, NF-κB reporter assays, and dose–response characterization in immune cell activation experiments. Endotoxin levels below 1.0 EU/μg eliminate LPS-driven artifacts that confound TNF-specific readouts in primary macrophage or dendritic cell cultures, supporting use in in vivo tumor biology models and as a reference standard in cytokine detection platforms such as ELISA. The C-terminal His-tagged construct, expressed in *E. coli* and purified to greater than 95% homogeneity, provides a suitable basis for antibody development workflows including immunogen preparation and positive-control validation.
