Description

This tag-free fragment spanning residues 32–65 of human PTH retains the biologically active N-terminal signaling domain critical for PTH1 receptor engagement and downstream cAMP generation. Validated in a cell-based assay, the protein induces cAMP accumulation in murine MC3T3E1 osteoblastic cells with an ED50 below 50 ng/ml, corresponding to a specific activity exceeding 2.0 × 10⁴ IU/mg—providing a reliable positive control for receptor binding assays, competition studies, and cell-based signaling experiments. Endotoxin levels below 1.0 EU/μg, combined with purity greater than 97% by SDS-PAGE, satisfy the criteria typically required for in vivo pharmacology models investigating calcium homeostasis or bone metabolism, as well as for use as a quantitative standard in immunoassay development. The E. coli-derived, tag-free format supports structure-function studies where post-translational modifications or fusion partners could confound receptor interaction analyses.
