TIGIT

TIGIT, the T-cell immunoglobulin and ITIM structural domain protein, alias WUCAM, Vstm3, VSIG9.TIGIT is encoded by the human TIGIT gene, which is located on chromosome 3 in the q13.33 region. TIGIT is an inhibitory receptor belonging to the Ig superfamily with an extracellular immunoglobulin structural domain, a type I transmembrane structure, and an intracellular tyrosine inhibition/activation motif for immunoreceptors (ITIM/ITAM).
The mechanism of action of TIGIT involves binding to ligands such as CD155, transmitting inhibitory signals through the intracellular ITIM motif, reducing the activation and proliferation of T cells and NK cells, and decreasing cytokine production.
TIGIT plays an important role in regulating the immune response of T cells and NK cells, especially in the tumor microenvironment, where high expression of TIGIT is associated with immunosuppression and tumor escape. TIGIT blockers have shown therapeutic potential in different tumor models and are novel targets for cancer therapy.
