ITGB6

Integrin beta-6 (ITGB6)​​, a member of the integrin β-subunit family, pairs exclusively with alpha-V (αV) to form the αVβ6 heterodimer. Primarily expressed in epithelial cells, this receptor binds latent TGF-β complexes and extracellular matrix proteins like fibronectin, driving TGF-β activation and cell-ECM interactions. ITGB6 overexpression is implicated in pathological fibrosis (e.g., idiopathic pulmonary fibrosis, liver cirrhosis) and epithelial-derived cancers (pancreatic, head/neck), where it promotes tissue remodeling, tumor invasion, and immune evasion. Its dysregulation often correlates with disease progression and poor clinical outcomes.
Therapeutic targeting of αVβ6 has gained momentum. Monoclonal antibodies such as ​​BG00011​​ (STX-100) block TGF-β activation and attenuate fibrosis in preclinical models, advancing to Phase II trials for pulmonary fibrosis. Small-molecule inhibitors and peptide-based agents are also under exploration to disrupt αVβ6-mediated cancer metastasis. Challenges include minimizing off-target effects due to αVβ6’s structural homology with other integrins. Recent advances in tissue-specific delivery systems and combination therapies with checkpoint inhibitors offer promising avenues to enhance efficacy while reducing systemic toxicity.
