ACVRL1

ACVRL1, also known as Activin Receptor-Like Kinase 1 (ALK1), is a crucial member of the Transforming Growth Factor β (TGF-β) superfamily. Structurally, it comprises a ligand-binding domain, a transmembrane region, and a serine/threonine kinase domain. ACVRL1 plays a pivotal role in endothelial cell differentiation, proliferation, and vascular development, serving as a positive regulator of angiogenesis. Upon ligand binding, ACVRL1 activates specific signaling pathways, such as the mitogen-activated protein kinase (MAPK) cascade, thereby regulating cellular processes like proliferation, differentiation, migration, and apoptosis. Additionally, ACVRL1 is involved in the activation of nuclear factor κB (NF-κB), crucial for modulating inflammatory and immune responses. However, mutations in the ACVRL1 gene are associated with diseases like hereditary hemorrhagic telangiectasia type 2 (HHT2), potentially leading to abnormal vascular development and increased vascular fragility.
