On April 25th each year, World Malaria Day is observed, aiming to raise global awareness about fighting malaria, increase knowledge about the disease, foster collaboration, and work towards a malaria-free world. Malaria, one of the deadliest infectious diseases worldwide, results in millions of deaths and illnesses annually, especially affecting children and pregnant women. This day commemorates the battle against malaria while promoting scientific advancements, strengthening prevention methods, and improving diagnosis and treatment. The theme of the event this year is "Accelerating the fight against malaria for a more equitable world".
Malaria is a deadly infection caused by parasites from the Plasmodium genus. Infected female Anopheles mosquitoes transmit these parasites when they bite humans, injecting sporozoites into the bloodstream. These sporozoites invade liver cells, grow into schizonts, and later attack red blood cells, causing recurring symptoms such as fever, chills, sweating, and headaches. Malaria types vary based on the Plasmodium species infecting humans, including tertian, quartan, ovale, and falciparum malaria.
Currently, malaria treatment mainly relies on targeted drug therapy depending on the malaria type, without requiring surgery. Due to increased resistance to conventional antimalarial drugs like chloroquine and sulfadoxine-pyrimethamine, researchers are searching for and verifying new drug targets. Special emphasis is placed on drugs targeting specific stages of the parasite's life cycle, e.g., those inhibiting liver-stage development or stopping parasite reproduction in red blood cells. Monoclonal antibody treatments have shown potential in recent years, effectively binding to and eliminating malaria parasites, thus curbing transmission and disease occurrence.
Malaria vaccine development remains a critical focus in global health. While the first malaria vaccine for children, RTS,S/AS01 (commercially known as Mosquirix), has been deployed in some high-risk regions, it offers limited and temporary protection. Therefore, scientists are exploring more potent and enduring vaccine strategies, including multi-epitope protein-based, RNA, and DNA vaccines.
