Images
  • IHC image of CSB-RA004928MA1HU diluted at 1:200 and staining in paraffin-embedded human tonsil tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-human polymer IgG labeled by HRP and visualized using 0.05% DAB.
Description

CD37 is a tetraspanin family member predominantly expressed on mature B cells, where it plays essential roles in B cell development, survival signaling, and immune cell interactions. This surface glycoprotein has emerged as a compelling target in hematological malignancy research, particularly in B cell lymphomas and chronic lymphocytic leukemia, making reliable detection tools invaluable for both basic immunology studies and translational research.

This recombinant monoclonal antibody, clone 9H6, offers the consistency and reproducibility that demanding research applications require. Generated against recombinant human CD37 protein and produced using recombinant technology, this antibody provides sequence-defined specificity with minimal lot-to-lot variation. The human IgG1 isotype format and affinity chromatography purification ensure high purity suitable for sensitive detection methods.

Validation in immunohistochemistry demonstrates reliable performance in formalin-fixed, paraffin-embedded human tissues. Testing in human tonsil tissue, a lymphoid organ rich in B cells, confirms specific staining at dilutions of 1:50 to 1:200 using citrate buffer antigen retrieval. The validation protocol employed a Leica Bond system with goat anti-human polymer detection, providing researchers with a well-characterized starting point for their own optimization.

Beyond immunohistochemistry, this antibody is also suitable for ELISA applications, offering flexibility across different experimental workflows. Whether investigating B cell biology, characterizing lymphoid tissue architecture, or exploring CD37 as a therapeutic target, this recombinant antibody delivers the reproducibility essential for generating publication-quality data in immunology and oncology research.

Target Background

Gene References into Functions
  1. CD37 is a critical determinant of rituximab plus cyclophosphamide, doxorubicin, vincristine, and prednisone outcome in diffuse large B-cell lymphoma. PMID:27760757
  2. Data indicate that cell differentiation antigen 37 (CD37) is well expressed and a potential drug target in acute myeloid leukemia (AML). PMID:25934707
  3. The CD37-targeted antibody-drug conjugate IMGN529 is highly active against human CLL and in a novel CD37 transgenic murine leukemia model. PMID:24445867
  4. Data indicate that enhanced antibody-dependent cell cytotoxicity (ADCC) is observed against chronic lymphocytic leukemia cells and is sustained at concentrations of SMIP-016(GV) as low at 5E(-6) microg/mL on cells expressing minimal CD37 antigen. PMID:23883821
  5. Identify two tyrosine residues in tetraspanin CD37 directly mediating transduction of survival and apoptotic signals. PMID:22624718
  6. In the absence of both CD37 and Tssc6, immune function is further altered when compared with CD37- or Tssc6-deficient transgenic mice, demonstrating a complementary role for these two molecules in cellular immunity. PMID:20709950
  7. CD37 cross-linking on human T cells transduces signals that lead to complete inhibition of CD3-induced T cell proliferation. PMID:14978098
  8. CD37 is important for dectin-1 stabilization in APC membranes and controls dectin-1-mediated IL-6 production. PMID:17182550
