Description
TSLP drives type 2 immune responses and epithelial–immune crosstalk in allergic inflammation, yet wild-type TSLP's heparin-binding properties can complicate quantitative assays; this R127A/R130A double mutant eliminates glycosaminoglycan interactions while preserving receptor engagement, enabling cleaner dose–response studies. Expressed as the full mature sequence (aa 29–159) in mammalian cells, the protein demonstrates specific antibody binding in functional ELISA with an EC50 of 52.55–58.67 ng/mL, supporting its use as a coating antigen in antibody development workflows, a positive control in TSLP detection assays, and a reference standard for validating anti-TSLP therapeutics. The endotoxin level below 1.0 EU/μg meets the contamination thresholds commonly required for primary human dendritic cell and T cell activation assays, where even trace LPS can trigger artifactual cytokine secretion and obscure TSLP-specific signaling through STAT5 and NF-κB pathways. Purity exceeding 95% by SDS-PAGE provides a suitable basis for quantitative studies of TSLP-driven Th2 polarization and ILC2 expansion.
