Description
The F176V polymorphism in FCGR3A confers higher affinity for IgG1 and IgG3 antibodies, making this variant essential for studies of antibody-dependent cellular cytotoxicity and therapeutic antibody efficacy in patient populations carrying this allele. Mammalian expression preserves the native glycosylation pattern critical for authentic Fc receptor interactions, while the extracellular domain spanning residues 17–208 captures the complete ligand-binding interface. Functional ELISA validation demonstrates dose-dependent binding to an anti-FCGR3A antibody with an EC50 of 7.8–12.2 ng/ml, confirming conformational integrity suitable for ligand-binding assays, therapeutic antibody epitope mapping, and competitive inhibition screening of biologics targeting the Fc-FcγRIIIa axis. The biotinylated C-terminus enables direct immobilization in SPR and BLI platforms for affinity characterization, while purity exceeding 95% and endotoxin below 1.0 EU/μg meet the quality thresholds commonly required for quantitative binding studies and antibody screening workflows.
