Description

IL-36γ is a pro-inflammatory IL-1 family cytokine that signals through IL-1RL2/IL-1RAcP to activate NF-κB and MAPK pathways, making it a key mediator in epithelial immune responses and psoriasis-related inflammation. This tag-free recombinant human IL-36G, covering the full mature sequence (aa 18–169), demonstrates robust signaling potency with an ED50 of 5–20 ng/mL in an IL-8 secretion assay using A431 human epithelial carcinoma cells, providing a reliable basis for cell-based activation assays, NF-κB and MAPK pathway dissection, and use as a reference standard in cytokine detection platforms. The protein's endotoxin level of less than 1.0 EU/μg satisfies the stringent criteria needed to prevent LPS-driven artifacts in sensitive immune cell experiments, including in vivo inflammation models and antibody development workflows such as ELISA coating or positive controls. Part of the IL-36 signaling system that is thought to be present in epithelial barriers and to take part in local inflammatory response; similar to the IL-1 system with which it shares the coreceptor IL1RAP. Seems to be involved in skin inflammatory response by acting on keratinocytes, dendritic cells and indirectly on T-cells to drive tissue infiltration, cell maturation and cell proliferation.
