Description
IL-12 drives Th1 polarization and NK cell activation through JAK2/TYK2-STAT4 signaling, making it a critical tool for dissecting innate-to-adaptive immune transitions. This tag-free human IL-12 heterodimer (IL12B aa 23–328 and IL12A aa 23–219) demonstrates exceptional potency with an ED50 of 0.01–0.05 ng/mL in PHA-stimulated human T lymphoblast proliferation assays, corresponding to a specific activity of approximately 1.1 × 10⁴ units/μg. The endotoxin level below 1.0 EU/μg satisfies the stringent criteria required to prevent LPS-driven artifacts in sensitive immune cell functional assays, including PBMC proliferation, Th1 differentiation studies, JAK/STAT pathway activation experiments, and in vivo tumor immunology models. Purity exceeding 95% by SDS-PAGE, combined with faithful representation of the mature heterodimer sequence, provides a suitable basis for use as a reference standard in cytokine detection assays or as a positive control in antibody validation workflows.
