IL-1 receptor antagonist protein (IL-1RN) is a critical endogenous regulator of IL-1 signaling, and this tag-free recombinant form covering the full mature sequence (aa 26–177) faithfully reproduces that antagonistic function with an ED50 of 13.2 ng/mL in an IL-1β–induced NF-κB reporter assay using 293-IL1R cells. This level of potency supports use in cell-based functional studies such as IL-1–dependent proliferation assays with TF-1 cells or PBMCs, NF-κB and MAPK pathway dissection, and in vivo inflammation models where precise dose–response control over IL-1 activity is essential. Endotoxin levels below 1.0 EU/μg minimize the risk of LPS-driven artifacts that can confound immune cell activation and cytokine readouts, which aligns with standards expected in primary cell culture and animal studies. Purity exceeding 95% by SDS-PAGE further provides a suitable basis for use as a reference standard in cytokine detection assays or as a positive control in ELISA-based antibody validation.
