Activity
Fully biologically active when compared to standard. rHusTRAIL-R2 reduced the production of LPS- induced TNF by its ability to neutralize endogenous TRAIL in fresh human PBMC. In this assay, endogenous TRAIL is induced during a 24 hour exposure to LPS (10 ng/mL) but in the presence of rHusTRAIL-R2, TRAIL-induced TNF is suppressed.

Description
TNFRSF10B (Death Receptor 5) serves as a critical decoy receptor for TRAIL, and this tag-free partial construct covering residues 52–183 captures the extracellular ligand-binding domain essential for TRAIL neutralization studies. Validated biological activity demonstrates that this protein suppresses LPS-induced TNF production in human PBMCs by neutralizing endogenous TRAIL over a 24-hour exposure, confirming functional ligand engagement suitable for competitive inhibition assays, blocking antibody screening, and TRAIL-pathway drug discovery. The E. coli expression system yields a properly folded ectodomain fragment that supports use in receptor-ligand interaction studies by SPR or BLI, epitope mapping for therapeutic antibody development, and as a positive control in binding assays. Purity exceeding 97% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based neutralization experiments and sensitive biophysical affinity characterization.
