Description
CCR4 is a seven-transmembrane GPCR notoriously difficult to produce as a full-length construct, and this recombinant human CCR4 (1–360 aa) overcomes that challenge through a cell-free E. coli expression system that enables recovery of the complete receptor sequence, including all transmembrane domains, with an N-terminal 10×His tag for oriented capture. Conformational integrity is supported by SPR data demonstrating high-affinity binding to an anti-CCR4 monoclonal antibody, with KD values of 3.16 nM (Biacore T200) and 1.71 nM (MetaSPR in FOS-12 detergent), while functional ELISA confirms dose-dependent interaction with an EC50 of 2.80–3.37 ng/mL. These validated binding characteristics provide a suitable basis for therapeutic antibody epitope mapping, competitive blocking antibody screening, affinity characterization by SPR or BLI, and small-molecule inhibitor discovery campaigns targeting CCR4-mediated signaling. Purity exceeds 85% by SDS-PAGE, which meets the quality thresholds commonly required for use as a positive control in receptor-ligand interaction assays.
