  • IHC image of CSB-RA092384A0HU diluted at 1:100 and staining in paraffin-embedded human kidney tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.
  • Immunofluorescence staining of U-251MG cell with CSB-RA092384A0HU at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
  • Overlay Peak curve showing SH-SY5Y cells stained with CSB-RA092384A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
Description

TOMM20 serves as a critical component of the translocase of the outer mitochondrial membrane complex, functioning as the primary receptor for nuclear-encoded mitochondrial preproteins. This receptor plays an essential role in mitochondrial protein import, making it an invaluable marker for studying mitochondrial dynamics, biogenesis, and cellular metabolism. Researchers investigating mitochondrial dysfunction in neurodegenerative diseases, cancer metabolism, and cellular stress responses frequently rely on TOMM20 detection to assess mitochondrial content and distribution.

This recombinant monoclonal antibody, clone 15F9, offers the reproducibility and consistency that demanding research applications require. Produced using recombinant technology with a defined sequence, this antibody eliminates the lot-to-lot variability that can compromise longitudinal studies and multi-site collaborations. The rabbit host and monoclonal nature ensure high specificity for human TOMM20, with affinity chromatography purification delivering a clean reagent suitable for sensitive detection methods.

Validation across multiple platforms demonstrates exceptional versatility for your experimental workflows. Western blot analysis confirms specific detection at the expected 16 kDa molecular weight across an extensive panel of human cell lines, including K562, THP-1, SH-SY5Y, U-251MG, Jurkat, HEK293, A431, HeLa, and PC-3 cells. Immunohistochemistry staining in human kidney tissue reveals clear mitochondrial localization patterns, while immunofluorescence in U-251MG cells and flow cytometry analysis in SH-SY5Y cells further demonstrate reliable performance in cell-based applications.

Whether you are quantifying mitochondrial mass, examining organelle distribution during cellular differentiation, or investigating mitochondrial dynamics in disease models, this antibody provides the consistent, validated performance essential for generating publication-quality data in mitochondrial biology research.

Target Background

Function(From Uniprot)
Central component of the receptor complex responsible for the recognition and translocation of cytosolically synthesized mitochondrial preproteins. Together with TOM22 functions as the transit peptide receptor at the surface of the mitochondrion outer membrane and facilitates the movement of preproteins into the TOM40 translocation pore. Required for the translocation across the mitochondrial outer membrane of cytochrome P450 monooxygenases.
Gene References into Functions
  1. TOMM20 and MCT1 were highly expressed in diffuse large B-cell lymphoma lymphocytes, indicating an OXPHOS phenotype, whereas non-neoplastic lymphocytes in the control samples did not express these markers. PMID:29248132
  2. TOMM20, MCT1, and MCT4 expression was significantly different in Hodgkin and Reed Sternberg (HRS) cells. HRS have high expression of TOMM20, while tumor associated macrophages have absent TOMM20 expression. Tumor-infiltrating lymphocytes have low TOMM20 expression. Reactive lymph nodes in contrast to cHL tumors had low TOMM20, MCT1, and MCT4 expression in lymphocytes and macrophages. PMID:29248133
  3. Gsk3beta and Tomm20 are substrates of the SCFFbxo7/PARK15 ubiquitin ligase associated with Parkinson's disease PMID:27503909
  4. TOMM20 protein is highly expressed in both anaplastic thyroid cancer and papillary thyroid cancer compared to non-cancerous thyroid tissue. PMID:26615136
  5. Tom20 was ubiquitously distributed in the organ of Corti, allowing well-delineated visualization of inner and outer hair cells. In the inner ear of aged subjects with Meniere's disease, there was decreased expression of Tom20. PMID:23165776
  6. a novel model in which Tom20 binds Tom70 to facilitate preprotein release from the chaperones by competition PMID:21771790
  7. Tom34 unlike Tom20 does not interact with the leader sequences of mitochondrial precursor proteins PMID:11913975
  8. the cytosolic domains of Tom20 and Tom22 function to maintain their substrate preproteins unfolded and prevent them from aggregating on the mitochondrial surface PMID:14699115
