Images
  • IHC image of CSB-RA243394A0HU diluted at 1:100 and staining in paraffin-embedded human stomach tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.
  • Overlay Peak curve showing jurkat cells stained with CSB-RA243394A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
Description

TAB1 (TGF-beta-activated kinase 1-binding protein 1) serves as a critical regulatory component in inflammatory and stress response signaling pathways. As an essential activator of TAK1 (MAP3K7), TAB1 plays a pivotal role in mediating signals downstream of TGF-beta, IL-1, and Toll-like receptors, making it a key focus for researchers investigating innate immunity, inflammatory diseases, and cellular stress responses.

This recombinant monoclonal antibody, clone 11B7, offers the reproducibility and consistency that demanding experimental workflows require. Developed using recombinant technology with a sequence-defined approach, this antibody eliminates the lot-to-lot variability that can compromise longitudinal studies or multi-site collaborations. The rabbit host origin combined with monoclonal specificity ensures reliable detection of human TAB1 across your experimental timeline.

Validation studies demonstrate robust performance in multiple applications, providing flexibility for diverse research needs. Immunohistochemistry testing in paraffin-embedded human stomach tissue using a Leica Bond system with citrate buffer antigen retrieval shows clear specific staining at 1:100 dilution, confirming suitability for tissue-based studies. Flow cytometry analysis in Jurkat cells reveals a distinct positive shift compared to isotype control, with fixed and permeabilized cells showing reliable intracellular detection at 1:100 dilution. These validated applications, along with ELISA compatibility, support comprehensive protein expression analysis from tissue architecture to single-cell resolution.

Supplied in a glycerol-containing buffer optimized for long-term stability, this antibody is well-suited for research programs exploring NF-κB signaling, inflammatory pathway regulation, and TAK1-mediated kinase cascades in human samples.

Target Background

Function(From Uniprot)
May be an important signaling intermediate between TGFB receptors and MAP3K7/TAK1. May play an important role in mammalian embryogenesis.
Gene References into Functions
  1. High TAB1 expression is associated with colorectal cancer. PMID:29328486
  2. TAB1 was identified as a functional target of miR-134, and the expression of TAB1 was increased by the transcription factors of NF-kappaB1, c-Rel, and ELK1 via miR-134. PMID:28206956
  3. we show that IL-1 induces robust p38a activation both in the nucleus and in the cytoplasm/membrane.Following stimulation, p38a activity returns to a basal level in absence of receptor degradation. While nuclear pulse is controlled by MKP1 through a negative feedback to pp38, its basal activity is controlled by both TAB1 and MKP1 through a positive feedback loop. PMID:27314954
  4. TAK1/TAB1 expression in non-small cell lung carcinoma tissue is significantly increased and closely associated with patient clinical prognosis. PMID:26884850
  5. miR-29a repressed TAB1-mediated TIMP-1 production in dermal fibroblasts, demonstrating that miR-29a may be a therapeutic target in SSc. PMID:25549087
  6. Data indicate that mitogen-activated protein kinase (MAPK) p38 activation is triggered by AMP-activated protein kinases (AMPK) and mediated by TAB1 protein. PMID:25151490
  7. The amino acid sequence between positions 373 and 502 of TAB1 is required for TP interaction. PMID:24462677
