Images
  • IHC image of CSB-RA011892MA1HU diluted at 1:50 and staining in paraffin-embedded human kidney tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Anti-Human lgG, Fcy Fragment Specific labeled by HRP and visualized using 0.05% DAB.
  • IHC image of CSB-RA011892MA1HU diluted at 1:50 and staining in paraffin-embedded human endometrial cancer performed on a Leica BondTM system.
  • Overlay Peak curve showing 786-O cells stained with CSB-RA011892MA1HU (red line) at 1:100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was Fluorescein (FITC) AffiniPure Goat Anti-Human IgG, Fcγ fragment specific at 1:200 dilution for 35min at 4℃.Control antibody (green line) was human IgG1 (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
Description

Integrin beta-8 plays a distinctive role among integrin family members, primarily functioning as a receptor for latent TGF-beta activation through its exclusive pairing with the alpha-v subunit. This specialized function positions ITGB8 as a critical regulator of TGF-beta bioavailability in tissues, with significant implications for developmental biology, fibrosis research, and tumor microenvironment studies. The protein's expression patterns in epithelial and neuronal tissues make it particularly relevant for investigators exploring tissue homeostasis and pathological remodeling processes.

This recombinant monoclonal antibody, clone 30G12, offers the reproducibility advantages inherent to sequence-defined production. Unlike traditional hybridoma-derived antibodies, recombinant manufacturing ensures consistent performance across lots, allowing researchers to establish reliable protocols without the variability that can complicate longitudinal studies or multi-site collaborations. The human IgG1 isotype format and affinity-chromatography purification contribute to a reagent suitable for demanding experimental workflows.

Validation studies demonstrate robust performance across multiple platforms. Immunohistochemistry applications have been confirmed in paraffin-embedded human kidney tissue and human endometrial cancer specimens at dilutions of 1:50 to 1:200, enabling detection in both normal and pathological contexts. Flow cytometry validation using 786-O renal carcinoma cells shows clear positive signal separation from isotype control, supporting cell surface detection studies at comparable dilutions.

This antibody serves researchers investigating integrin-mediated TGF-beta activation, epithelial biology, and cancer progression. The demonstrated performance in both tissue sections and live cell analysis provides flexibility for studies examining ITGB8 expression patterns, from archival clinical specimens to functional cell-based assays exploring integrin biology in disease contexts.

Target Background

Function(From Uniprot)
Integrin alpha-V:beta-8 (ITGAV:ITGB8) is a receptor for fibronectin. It recognizes the sequence R-G-D in its ligands. Integrin alpha-V:beta-6 (ITGAV:ITGB6) mediates R-G-D-dependent release of transforming growth factor beta-1 (TGF-beta-1) from regulatory Latency-associated peptide (LAP), thereby playing a key role in TGF-beta-1 activation on the surface of activated regulatory T-cells (Tregs) (Probable). Required during vasculogenesis.
