Description
ERBB2 (HER2) is a critical oncogenic receptor tyrosine kinase whose extracellular domain serves as the primary target for therapeutic antibodies in HER2-positive cancers, making conformationally accurate recombinant protein essential for meaningful binding studies. This construct spans the extracellular region (aa 23–652) with a C-terminal 6xHis tag, and its mammalian cell expression preserves the native glycosylation and disulfide bonding required for proper domain folding and epitope presentation. Functional ELISA confirms that immobilized HER2 at 2 μg/ml binds Trastuzumab with an EC50 of 2.179–2.825 ng/ml, directly supporting applications in therapeutic antibody epitope mapping, competitive blocking antibody screening, and affinity characterization by SPR or BLI.
