Folate receptor alpha mediates cellular uptake of folate and folate-conjugated therapeutics, making it a validated target in oncology and antibody-drug conjugate development. This mammalian-expressed construct spanning residues 25–233 captures the extracellular ligand-binding domain with native glycosylation patterns that preserve conformational epitopes critical for therapeutic antibody screening and epitope mapping studies. Functional ELISA validation demonstrates specific antibody binding with an EC50 of 6.893–7.883 ng/mL, confirming that the protein retains native structure suitable for competitive inhibition assays, blocking antibody screening, and affinity characterization by surface plasmon resonance or biolayer interferometry. The combination of >95% purity and <1.0 EU/μg endotoxin meets the quality thresholds commonly required for ligand-binding assays, small-molecule inhibitor screening, and use as a positive control in receptor-ligand interaction studies targeting folate pathway biology.
