Description

ZNRF3 functions as a critical negative regulator of Wnt signaling by targeting Frizzled receptors for ubiquitin-dependent degradation, making it a key node for studying Wnt pathway modulation. This recombinant human ZNRF3 partial construct (aa 56–219), expressed in mammalian cells with a C-terminal 6xHis tag, demonstrates confirmed binding to human RSPO2 with an EC50 of 5.091–6.991 ng/mL in functional ELISA, supporting its use as a positive control in ligand-binding assays, inhibitor screening for IC50 determination, and R-spondin interaction studies. The tag placement at the C-terminus preserves the extracellular domain architecture required for accurate kinetic parameter analysis and substrate specificity profiling.
