Description
ASGR1 mediates the endocytosis of desialylated glycoproteins in hepatocytes, a pathway exploited for liver-targeted drug delivery and implicated in metabolic regulation and hepatocellular carcinoma progression. This full-length construct (aa 1–291) expressed in mammalian cells preserves native glycosylation and disulfide architecture, as evidenced by sub-nanomolar affinity (KD = 0.716 nM) to a validated monoclonal antibody measured by surface plasmon resonance and an EC50 of 2.715–3.605 ng/mL in functional ELISA. The demonstrated binding kinetics support use in therapeutic antibody epitope mapping, competitive inhibition screening for liver-targeting biologics, and ligand-receptor interaction studies by SPR or biolayer interferometry. With purity exceeding 85% by SDS-PAGE and endotoxin below 1.0 EU/μg, this protein aligns with standards expected in antibody validation workflows and small-molecule inhibitor discovery campaigns targeting hepatic lectin pathways.
