Description
CD73 (NT5E) serves as a critical ectoenzyme in purinergic signaling, hydrolyzing extracellular AMP to adenosine and thereby modulating immune responses within the tumor microenvironment. This recombinant human CD73 encompasses the full mature protein (aa 27–549) expressed in mammalian cells with a C-terminal 6×His tag, positioning the tag away from the catalytic domain to preserve native enzymatic function. Binding activity has been confirmed by functional ELISA, where immobilized CD73 at 2 μg/mL bound an anti-CD73 monoclonal antibody with an EC50 of 3.212–4.525 ng/mL, supporting its use as a positive control in enzyme-linked assays, inhibitor screening for IC50 determination, and kinetic parameter analysis (Km, Vmax, kcat). Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typically required for accurate specific activity measurements, substrate specificity profiling, and drug candidate evaluation studies targeting adenosinergic pathways.
