Activity
Malic Enzyme activity was assayed spectrophotometrically at 340nm as described in Mandela and Sauer (1975). The standard reaction mixture contained 50 mM Tris-HCl, 3 mM MnCl2, 5 mM malate, 0.12 mM NADP+, 2.5 mM fumarate. Assay was performed in a Beckman spectrophotometer. The Km value is 1.5 ± 0.6 mM

Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.

Description
Mitochondrial NAD-dependent malic enzyme (ME2) occupies a critical metabolic node linking the TCA cycle to NADPH production, making it an emerging target in cancer metabolism research. This recombinant human ME2 (residues 19–584, full-length mature protein) carries a C-terminal 6xHis tag positioned away from the catalytic domain, preserving native enzymatic function as confirmed by a validated Km of 1.5 ± 0.6 mM for malate under spectrophotometric assay conditions (340 nm, Tris-HCl/MnCl₂/fumarate activation system). The measured kinetic parameters support direct use in substrate specificity profiling, Vmax and kcat determination, inhibitor screening with IC50 quantification, and drug candidate mechanism-of-inhibition studies. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the stringent criteria required for accurate kinetic measurements, positive-control applications in enzyme-linked assays, and structural studies such as crystallography.
