Thyroid peroxidase (TPO) is a key enzyme in thyroid hormone biosynthesis, catalyzing the iodination of tyrosine residues in thyroglobulin and the coupling of iodotyrosines to form thyroxine (T4) and triiodothyronine (T3). TPO sits in the apical membrane of thyroid follicular cells and is required for normal thyroid function and hormone production. TPO serves as a major autoantigen in autoimmune thyroid diseases. Anti-TPO antibodies are important diagnostic markers for conditions such as Hashimoto's thyroiditis and Graves' disease.
