Description

This ELISA kit provides quantitative measurement of human SPAM1 (sperm adhesion molecule 1/hyaluronidase PH-20) in research contexts spanning signal transduction and reproductive biology. Because SPAM1 is a GPI-anchored hyaluronidase that can be released from cell surfaces into conditioned media, detecting its soluble form at low concentrations demands purpose-built assay sensitivity.

Detection Range: The 78–5000 pg/mL range captures the low-to-moderate concentrations of shed SPAM1 typically found in cell culture supernates from transfected lines or primary reproductive tract cells, minimizing the need for extensive sample concentration while accommodating higher-expression systems through simple dilution.

Sensitivity: At 39 pg/mL, the kit detects SPAM1 in conditioned media from cells with modest endogenous expression, including epididymal or cumulus cell cultures where soluble PH-20 levels may be near the lower physiological threshold.

Sample Compatibility: Validation for cell culture supernates directly supports in vitro studies of SPAM1 shedding mechanisms, GPI-anchor cleavage regulation, and hyaluronidase activity modulation without requiring cell lysis or membrane fractionation.

Involved in sperm-egg adhesion. Upon fertilization sperm must first penetrate a layer of cumulus cells that surrounds the egg before reaching the zona pellucida. The cumulus cells are embedded in a matrix containing hyaluronic acid which is formed prior to ovulation. This protein aids in penetrating the layer of cumulus cells by digesting hyaluronic acid.
