TGFA

Transforming Growth Factor Alpha (TGFA), also known as TGF-α, is a mitogenic polypeptide that functions as a ligand for the epidermal growth factor receptor (EGFR). It activates downstream pathways including MAPK/ERK and PI3K/AKT through receptor dimerization and tyrosine kinase phosphorylation, promoting epithelial cell proliferation, differentiation, and survival. TGFA plays critical roles in embryonic development, wound healing, and tissue regeneration. Its expression is tightly regulated under physiological conditions but becomes dysregulated in multiple pathological contexts, particularly in cancers characterized by EGFR hyperactivation.
Overexpression of TGFA correlates with tumorigenesis in colorectal, lung, and hepatocellular carcinomas through sustained EGFR signaling. It also contributes to inflammatory skin disorders such as psoriasis via aberrant keratinocyte activation. Therapeutic strategies targeting TGFA-associated pathways include EGFR monoclonal antibodies (e.g., cetuximab) and tyrosine kinase inhibitors (erlotinib, gefitinib), though resistance mechanisms remain challenging. Recent trials explore combination therapies with MEK or STAT3 inhibitors to overcome resistance. Diagnostic applications leverage TGFA as a biomarker for EGFR-driven malignancies and therapeutic response monitoring.
