PRKACA, cAMP-dependent protein kinase catalytic subunit alpha, also known as PKCA1, Cs-PKA. PRKACA is the alpha catalytic subunit of protein kinase A (PKA), a cAMP-activated serine/threonine protein kinase that consists of two catalytic subunits and two regulatory subunits. In the absence of cAMP, PKA exists as a blunted complex. cAMP binds to the regulatory subunits, resulting in a conformational change of the regulatory subunits that dissociates the regulatory subunits from the catalytic subunits, releasing the catalytic subunits, which in turn phosphorylates serine or threonine residues of certain proteins in the cell, altering the activity of these proteins and affecting the expression of related genes.
PRKACA is involved in the regulation of a variety of cellular functions, including the metabolism of glycogen, sugars, and lipids, as well as physiological processes such as the circadian sleep/wake cycle and rapid eye movement (REM) sleep. Abnormal PRKACA activity has been associated with a variety of diseases, such as adrenocortical adenomas, congestive heart failure, and primary pigmented nodular adrenocortical disorders.
