NECTIN4

NECTIN4, or connexin cell adhesion molecule 4, alias LNIR, PRR4, EDSS1, PVRL4. NECTIN4 protein is a transmembrane cell adhesion molecule belonging to the Nectins family of immunoglobulin-like molecules with an extracellular structural region consisting of an IGV loop and two IGC loops, a transmembrane structural domain, and a cytoplasmic structural domain that contains an afadin binding motif.
NECTIN4 is involved in intercellular adhesion through interactions with other Nectins molecules, forming physical junctions between neighboring cells that are essential for enabling intercellular communication, migration, and other important cellular processes.
NECTIN4 is overexpressed in a variety of tumors, such as uroepithelial carcinoma, breast cancer, and pancreatic cancer, while its expression in normal tissues is low. Overexpression of NECTIN4 is associated with tumor cell proliferation, differentiation, migration, invasion and other processes, and promotes tumor cell proliferation and migration mainly through activation of the PI3K/Akt pathway, which is a potential target for tumor therapy, especially an important target in the therapeutic strategy of antibody-drug-coupled compounds (ADCs).
