LILRB2

LILRB2, a member of the leukocyte immunoglobulin-like receptor subfamily B, is an immune-suppressive type I transmembrane glycoprotein. Structurally, it features typical immunoglobulin-like extracellular domains, a transmembrane region, and intracellular immunoreceptor tyrosine-based inhibitory motifs (ITIMs). Functionally, LILRB2 regulates immune responses, maintains immune tolerance, and influences cell differentiation by recognizing and binding to various ligands, such as HLA-G and ANGPTLs. Notably, in the context of cancer, LILRB2 interacts with ligands in the tumor microenvironment, promoting tumor growth and facilitating immune evasion. Upon ligand binding, LILRB2 initiates a cascade of signaling events, including the phosphorylation of ITIMs and subsequent activation of downstream signaling molecules, thereby modulating the activity and function of immune cells. Further elucidating the structure, function, and signaling pathways of LILRB2 holds significant promise for developing novel therapeutic strategies in cancer treatment.
