LAG3

LAG-3 is an immunoinhibitory transmembrane protein comprised of key components, including four immunoglobulin superfamily domains in the extracellular region, a transmembrane domain, and serine phosphorylation sites in the intracellular domain. Functionally, LAG-3 primarily interacts with ligands such as MHC II and galectin-3 to suppress T cell activation and cytokine secretion, thereby maintaining immune homeostasis. In terms of signaling, LAG-3 engagement with its ligands triggers intracellular interactions with downstream signaling molecules, modulating the activation state of T cells. In cancer immunotherapy, LAG-3 serves as a crucial target, and blocking or inhibiting LAG-3 can potentiate T cell-mediated tumor killing. Therefore, elucidating the structure, function, and signaling pathways of LAG-3 holds significant promise for the development of novel immunotherapy strategies.
