COL18A1, a significant member of the collagen family, possesses a distinct molecular structure that plays a pivotal role in the extracellular matrix through its specific amino acid sequence. Functionally, COL18A1 is not only involved in the formation and maintenance of the retina, ensuring its normal function, but also regulates the migration, proliferation, and adhesion of endothelial cells, crucial for processes such as angiogenesis, wound healing, and placental circulation establishment. Additionally, COL18A1 specifically inhibits endothelial cell proliferation and angiogenesis by suppressing endothelial cell migration and adhesion, as well as downregulating the expression of angiogenesis-related genes. In terms of signaling, COL18A1 triggers a series of intracellular signal transductions through binding to endothelial cell surface receptors, involving interactions with molecules such as transglutaminase 2, heparin, integrins α5 beta 1, and αV beta 3.
