CD3G

CD3G encodes the gamma subunit of the CD3 complex, a critical component of the T-cell receptor (TCR). The protein, officially named T-cell surface glycoprotein CD3 gamma chain (CD3γ), is also known as T3G or CD3γ. It associates with CD3δ, ε, and ζ chains to form the TCR-CD3 complex, essential for TCR surface expression and signal transduction. Upon antigen recognition, CD3γ cytoplasmic immunoreceptor tyrosine-based activation motifs (ITAMs) undergo phosphorylation, initiating downstream signaling through ZAP70, LCK, and LAT adaptors. This activates NF-κB, MAPK, and calcium-NFAT pathways, driving T-cell proliferation, differentiation, and cytokine production.
CD3γ dysfunction is linked to severe combined immunodeficiency (SCID) and autoimmune disorders like lupus. Aberrant TCR signaling contributes to T-cell lymphomas and resistance in checkpoint inhibitor therapies. Therapeutic strategies targeting CD3 include anti-CD3 monoclonal antibodies (e.g., teplizumab for type 1 diabetes) and bispecific T-cell engagers (e.g., blinatumomab for B-cell malignancies). Emerging CAR-T platforms optimize CD3ζ-derived signaling domains to enhance antitumor efficacy. A phase II trial (NCT04503278) is evaluating CD3γ-specific nanobodies for autoimmune modulation.
