Description
FAS (CD95) serves as the critical death receptor initiating extrinsic apoptosis upon engagement with Fas Ligand, making it a central target in cancer immunology and therapeutic antibody development. This tag-free recombinant human FAS encompasses the extracellular ligand-binding domain (aa 17–173), and its biological activity is confirmed by an ED50 of 10–15 µg/ml for inhibiting FasL-induced cytotoxicity in Jurkat cells, directly validating its use in competitive inhibition assays, blocking antibody screening, and receptor-ligand interaction studies via ELISA or SPR. The E. coli-expressed protein achieves >95% purity by SDS-PAGE with endotoxin levels below 1.0 EU/µg, satisfying the quality criteria typical for cell-based functional assays and drug discovery screens targeting FAS/FasL pathway modulation. Researchers can confidently employ this protein as a positive control in binding assays or for epitope mapping of therapeutic antibody candidates directed against the FAS ectodomain.
