Description
TNFRSF12A (Fn14/TweakR) serves as the sole receptor for TWEAK, making it a critical node in tumor microenvironment signaling, angiogenesis, and NF-κB pathway activation. This tag-free recombinant construct spans the extracellular cysteine-rich domain (residues 28–80), providing the complete ligand-binding region validated by functional inhibition of TWEAK-dependent HUVEC proliferation with an ED50 below 30 ng/mL (specific activity >3.3 × 10⁴ IU/mg in the presence of 15 ng/mL rHuTWEAK). These activity data support use as a competitive inhibitor in blocking antibody screening, as a positive control in TWEAK–Fn14 binding assays (ELISA, SPR, BLI), and for small-molecule or biologic inhibitor discovery targeting this axis. Expressed in *E. coli* without a fusion tag, the protein provides >95% purity by SDS-PAGE and endotoxin levels below 1.0 EU/μg, satisfying the quality criteria typical for cell-based proliferation assays and ligand-receptor interaction studies.
