Description
TNFRSF11A (RANK) serves as the essential receptor for RANKL-mediated osteoclast differentiation and NF-κB signaling, making its soluble extracellular domain a critical tool for dissecting RANK–RANKL interactions in oncology research. This tag-free recombinant construct spanning residues 29–202 demonstrates potent ligand-neutralizing activity, with an ED50 below 50 ng/mL in inhibiting sRANKL-induced NF-κB activation in RAW 264.7 cells, corresponding to a specific activity exceeding 2.0 × 10⁴ IU/mg. These functional data support use in competitive inhibition assays, blocking antibody screening, RANKL-neutralization studies, and as a positive control in SPR or BLI-based ligand-binding experiments. Purity exceeding 98% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based functional assays and therapeutic antibody epitope mapping workflows.
