Claudin-6 functions as a tight junction protein with emerging significance in cancer biology, where its aberrant expression marks certain tumor types and positions it as a therapeutic target. This full-length construct (aa 1–219) displays quantifiable binding to anti-CLDN6/9 recombinant antibodies in functional ELISA, with EC50 values of 4.027–6.270 ng/mL and 7.349–12.12 ng/mL depending on antibody format, demonstrating that the protein adopts a conformation recognized by therapeutic-relevant binders. These binding kinetics support use in antibody development and validation workflows, particularly for screening candidate therapeutics against native epitopes, and provide a suitable basis for cell adhesion assays investigating claudin-mediated junction dynamics in tumor models. Mammalian expression yields purity greater than 95% by SEC-HPLC and endotoxin below 1.0 EU/μg, meeting the quality thresholds commonly required for cell-based migration and invasion studies where low endotoxin is critical to avoid confounding inflammatory responses.
