  • Immunofluorescence staining of HepG2 cell with CSB-RA250248A0HU at 1:50, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
  • Overlay Peak curve showing Hela cells stained with CSB-RA250248A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10,000 events was performed.
Description

Keratin 16 is a type I intermediate filament protein that plays a critical role in epithelial cell structure and stress response. Expressed primarily in stratified epithelia, KRT16 serves as a valuable marker for studying keratinocyte differentiation, wound healing, and inflammatory skin conditions. Its upregulation in hyperproliferative states makes it particularly relevant for researchers investigating psoriasis, wound repair mechanisms, and epithelial cancers.

This recombinant monoclonal antibody, generated against a synthetic peptide derived from human KRT16, offers the reproducibility and consistency that demanding experimental workflows require. Because the antibody sequence is defined and produced recombinantly, you can expect reliable performance across experiments and over time, eliminating the lot-to-lot variability that can complicate long-term studies or multi-site collaborations.

Validation testing demonstrates robust performance across multiple applications, giving you flexibility in your experimental design. Western blot analysis of MCF-7 whole cell lysate reveals a clean band at the predicted molecular weight of 51 kDa, confirming specific target recognition. For cellular localization studies, immunofluorescence staining has been validated in HepG2 cells, while flow cytometry analysis in HeLa cells shows clear positive signal separation from isotype controls, enabling quantitative assessment of KRT16 expression at the single-cell level.

Whether you are characterizing epithelial differentiation programs, investigating keratin expression patterns in cancer cell lines, or exploring the molecular basis of skin disorders, this antibody provides a dependable tool for your research. The validated performance across Western blot, immunofluorescence, and flow cytometry applications supports diverse experimental approaches within a single, well-characterized reagent.

Target Background

Function(From Uniprot)
Epidermis-specific type I keratin that plays a key role in skin. Acts as a regulator of innate immunity in response to skin barrier breach: required for some inflammatory checkpoint for the skin barrier maintenance.
Gene References into Functions
  1. Study identified iRHOM2 as a novel regulator of K16 in humans and mice, with important implications for palmoplantar keratodermas, wound healing, inflammatory skin disease and cancers. PMID:28128203
  2. results suggest a conditional regulation of KRT16 gene by ATF4 that may be inhibited in normal cells, but engaged during cancer progression. Potential roles of KRT16, FAM129A and HKDC1 genes upregulation in adaptive stress responses and pathologies are discussed PMID:29420561

  4. The wider spectrum of KRT16 mutations suggests that changes in codons 125, 127, and 132 are most commonly responsible for PC-1 and that proline substitution mutations at codons 127 or 128 may produce more severe disease PMID:24357266

  8. Patients with p.Asn125Asp and p.Arg127Pro mutations in KRT16 exhibited more severe disease than patients carrying p.Asn125Ser and p.Arg127Cys mutations in terms of age of onset of symptoms, extent of nail involvement, and impact on daily quality of life. PMID:21160496
  9. Data suggest that an immune response to trichohyalin and K16 may have a role in the pathogenesis of the enigmatic disorder. PMID:20722389
