Images
  • IHC image of CSB-RA310122A0HU diluted at 1:100 and staining in paraffin-embedded human kidney tissue performed on a Leica BondTM system. After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB.
  • IHC image of CSB-RA310122A0HU diluted at 1:100 and staining in paraffin-embedded human colorectal cancer performed on a Leica BondTM system.
  • Overlay Peak curve showing 786-O cells stained with CSB-RA310122A0HU (red line) at 1:100. The cells were fixed in 4% formaldehyde and permeated by 0.2% TritonX-100 for 10min. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*106cells) for 45min at 4℃. The secondary antibody used was FITC-conjugated goat anti-rabbit IgG (H+L) at 1/200 dilution for 35min at 4℃.Control antibody (green line) was Rabbit IgG (1ug/1*106cells) used under the same conditions. Acquisition of >10, 000 events was performed.
Description

BNIP3L, also known as NIX, is a BH3-only protein that plays a central role in programmed mitochondrial clearance through mitophagy. This protein is essential for the selective elimination of mitochondria during erythrocyte maturation and serves as a critical regulator of cell death pathways. BNIP3L has garnered significant research attention for its involvement in hypoxia-induced autophagy, cardiac homeostasis, and tumor suppression, making it a valuable target for studies spanning cancer biology, metabolic regulation, and developmental processes.

This recombinant monoclonal antibody, clone 12C4, offers the consistency and reliability that demanding experimental workflows require. Produced using recombinant technology in rabbit host, this antibody provides sequence-defined specificity against human BNIP3L, ensuring reproducible results across experiments and eliminating the lot-to-lot variability often encountered with traditional hybridoma-derived antibodies. The affinity-chromatography purification yields a highly pure reagent suitable for sensitive detection applications.

Validation studies demonstrate robust performance across multiple platforms. Immunohistochemistry staining in paraffin-embedded human kidney tissue and colorectal cancer specimens at 1:100 dilution reveals clear target detection using standard citrate buffer antigen retrieval protocols on automated Leica Bond systems. Flow cytometry analysis using the 786-O renal cell carcinoma line confirms intracellular detection capability, with distinct signal separation from isotype controls when cells are fixed and permeabilized.

Whether investigating mitophagy mechanisms, exploring BNIP3L's role in tumor microenvironment adaptation, or examining its function in tissue-specific contexts, this antibody provides the experimental flexibility researchers need. The validated performance in both tissue sections and cell-based assays supports diverse cell biology research applications.

Target Background

Function(From Uniprot)
Induces apoptosis. Interacts with viral and cellular anti-apoptosis proteins. Can overcome the suppressors BCL-2 and BCL-XL, although high levels of BCL-XL expression will inhibit apoptosis. Inhibits apoptosis induced by BNIP3. Involved in mitochondrial quality control via its interaction with SPATA18/MIEAP: in response to mitochondrial damage, participates in mitochondrial protein catabolic process (also named MALM) leading to the degradation of damaged proteins inside mitochondria. The physical interaction of SPATA18/MIEAP, BNIP3 and BNIP3L/NIX at the mitochondrial outer membrane regulates the opening of a pore in the mitochondrial double membrane in order to mediate the translocation of lysosomal proteins from the cytoplasm to the mitochondrial matrix. May function as a tumor suppressor.
Gene References into Functions
  1. The results demonstrate that Nix can serve as an alternative mediator of mitophagy to maintain mitochondrial turnover, identifying Nix as a promising target for neuroprotective treatment in PINK1/Parkin-related Parkinson's disease. PMID:28281653
  2. Here the authors present evidence for phosphorylation-driven regulation of the Nix:LC3B interaction. Isothermal titration calorimetry and NMR indicate a ~100 fold enhanced affinity of the serine 34/35-phosphorylated Nix LC3-interacting region (LIR) to LC3B and formation of a very rigid complex compared to the non-phosphorylated sequence. PMID:28442745
  3. p75(NTR) and NIX may play critical roles in intracerebral hemorrhage-induced neuronal apoptosis in vitro and in vivo. PMID:27726026
  4. Data suggest that targeting BNIP3L protein in wild-type p53 tumor suppressor colon cancer cells is an anticancer strategy activating iron depletion signaling and the mitophagy-related cell death pathway. PMID:26517689
  5. regulates mitophagy during hypoxia, whereas NIX is required for mitophagy during development of the erythroid lineage. PMID:25753537

  12. NIX is required for the selective elimination of mitochondria, however, because mitochondrial clearance, in the absence of NIX, is arrested at the stage of mitochondrial incorporation into autophagosomes and autophagosome maturation. PMID:18048346
