Description

BIRC3, also known as cellular inhibitor of apoptosis 2 (c-IAP2), plays a critical role in regulating programmed cell death and NF-κB signaling pathways. As a RING-type E3 ubiquitin ligase, BIRC3 modulates cell survival decisions by inhibiting caspases and facilitating ubiquitin-dependent signaling downstream of TNF receptor complexes. Its dysregulation has been implicated in various malignancies and inflammatory conditions, making it a significant target for researchers investigating apoptotic mechanisms, cancer biology, and immune signaling networks.

This recombinant rabbit monoclonal antibody (clone 7E9) offers the reproducibility and consistency that demanding experimental workflows require. Generated against a synthetic peptide derived from human BIRC3, the antibody is produced using recombinant technology, ensuring sequence-defined specificity and eliminating the lot-to-lot variability that can compromise longitudinal studies. Affinity chromatography purification delivers a reagent suitable for quantitative applications where signal reliability matters.

Validation studies confirm robust performance in flow cytometry applications, with recommended dilutions of 1:50 to 1:200. Flow cytometric analysis using Raji cells demonstrated clear positive signal separation from isotype controls, with intracellular staining performed following fixation and permeabilization. Western blot analysis in Raji whole cell lysate detected a band at approximately 72 kDa, slightly higher than the predicted 68 kDa molecular weight—a shift commonly attributable to post-translational modifications such as ubiquitination or phosphorylation events consistent with BIRC3's regulatory functions.

This antibody serves researchers studying apoptosis regulation, NF-κB pathway dynamics, and ubiquitin-mediated signaling in human samples, providing a reliable tool for both cell-based assays and protein detection applications.

Target Background

Function(From Uniprot)
Multi-functional protein which regulates not only caspases and apoptosis, but also modulates inflammatory signaling and immunity, mitogenic kinase signaling and cell proliferation, as well as cell invasion and metastasis. Acts as an E3 ubiquitin-protein ligase regulating NF-kappa-B signaling and regulates both canonical and non-canonical NF-kappa-B signaling by acting in opposite directions: acts as a positive regulator of the canonical pathway and suppresses constitutive activation of non-canonical NF-kappa-B signaling. The target proteins for its E3 ubiquitin-protein ligase activity include: RIPK1, RIPK2, RIPK3, RIPK4, CASP3, CASP7, CASP8, IKBKE, TRAF1, and BCL10. Acts as an important regulator of innate immune signaling via regulation of Toll-like receptors (TLRs), Nodlike receptors (NLRs) and RIG-I like receptors (RLRs), collectively referred to as pattern recognition receptors (PRRs). Protects cells from spontaneous formation of the ripoptosome, a large multi-protein complex that has the capability to kill cancer cells in a caspase-dependent and caspase-independent manner. Suppresses ripoptosome formation by ubiquitinating RIPK1 and CASP8.
