Target Background

Function(From Uniprot)
Involved in chromosome cohesion during cell cycle and in DNA repair. Central component of cohesin complex. The cohesin complex is required for the cohesion of sister chromatids after DNA replication. The cohesin complex apparently forms a large proteinaceous ring within which sister chromatids can be trapped. At anaphase, the complex is cleaved and dissociates from chromatin, allowing sister chromatids to segregate.
Gene References into Functions
  1. The X. laevis Eco2 bound to its primary and secondary Smc3 substrates demonstrate the plasticity of the substrate-binding site, which confers substrate specificity by concerted conformational changes of the central beta-hairpin and the C-terminal extension. PMID:28290497
  2. Smc3 deacetylation by Hos1, triggered by Scc1 cleavage, facilitates efficient dissolution of sister chromatid cohesion during early metaphase. PMID:29100057
  3. Sister chromatid cohesion is mediated by entrapment of sister DNAs by a tripartite ring composed of cohesin's Smc1, Smc3, and alpha-kleisin subunits; the function of Smc3 acetylation is to block its dissociation from alpha-kleisin PMID:22901742
  4. Data provide evidence that Smc3p acetylation, which has an essential role in cohesion establishment, modulates the Smc3p ATP-bound state. PMID:20498298
  5. Hos1 deacetylates Smc3 to close the cohesin acetylation cycle. PMID:20832720
  6. A cycle of Smc3 NBD acetylation is therefore an essential aspect of the chromosome cycle in eukaryotic cells PMID:20832721

Subcellular Location
Nucleus. Chromosome. Note=Associates with chromatin. Before prophase it is scattered along chromosome arms. At anaphase, the MCD1 subunit of the cohesin complex is cleaved, leading to the dissociation of the complex from chromosomes, allowing chromosome separation.
Protein Families
SMC family, SMC3 subfamily
