Target Background

Function(From Uniprot)
Dual-specificity kinase which possesses both serine/threonine and tyrosine kinase activities. May play a role in a signaling pathway regulating nuclear functions of cell proliferation. Modulates alternative splicing by phosphorylating the splice factor SRSF6. Exhibits a substrate preference for proline at position P+1 and arginine at position P-3. Has pro-survival function and negatively regulates the apoptotic process. Promotes cell survival upon genotoxic stress through phosphorylation of SIRT1. This in turn inhibits TP53 activity and apoptosis.
Gene References into Functions
  1. Dyrk1A involved in the regulation of alternative splicing of tau exon 10 in vivo. Overexpression of Dyrk1A in neonatal Ts65Dn mouse brains suppressed tau exon 10 inclusion, leading to an increase of 3R-tau/4R-tau ratio. Inhibition of Dyrk1A promotes endogenous tau exon 10 inclusion in cultured cells and in neonatal rat brain. PMID:28377597
  2. Identify Dyrk1a as a novel negative regulator of D-cyclin-mediated Rb1/E2f-signalling. As dysregulation of this pathway with impaired cardiomyocyte proliferation leads to cardiomyopathy. PMID:27056896
  3. Enhanced activation of Dyrk1A-ASF-CaMKIIdelta signaling pathway may underlie the mechanisms of HF after MI, and Dyrk1A inhibition may contribute to inactivation of this pathway and thereby retard the progression of MI-induced HF. PMID:26067684
  4. calcineurin/NFAT pathway mediates the upregulation of PAI-1 by NGF PMID:23825664
  5. A model of DYRK autoactivation has been proposed in which tyrosine autophosphorylation in the activation loop stabilizes a conformation of the catalytic domain with enhanced serine/threonine kinase activity without disabling tyrosine phosphorylation. PMID:23809146
  6. Dyrk1A negatively regulates actin filament assembly by phosphorylating N-WASP, which ultimately promotes the intramolecular interaction of its GTPase-binding and verprolin, cofilin and acidic domains PMID:22250195
  7. Phosphorylation of SRp55 by Dyrk1A suppressed its ability to promote Tau exon 10 inclusion. PMID:22767602
  8. Decreased expression of Dyrk1a in rat heart leads to cardiomyocyte hypertrophy in hyperhomocysteinemic rats. PMID:19906449
  9. In cultured cardiomyocytes, adenovirus-mediated overexpression of DYRK1A antagonized calcineurin-mediated nuclear NFAT translocation and the phenylephrine-induced hypertrophic growth response. PMID:21273244
  10. the regulation of DYRK1A by REST in a negative feedback loop suggests that DYRK1A and REST are closely related in neurodevelopment. PMID:21252229
  11. The result suggests that Mnbk/Dyrk1A can play a dual role in regulating the interaction of dynamin 1 with amphiphysin 1 PMID:11877424
  12. DYRK1A blunted cardiomyocyte hypertrophy induced by overexpression of constitutively active calcineurin and attenuated the induction of the hypertrophic gene program. PMID:19372220
  13. DYRK1A attenuates Notch signalling in neural cells, constituting a novel mechanism to modulate different developmental processes and contribute to the alterations observed during brain development in animal models of Down syndrome. PMID:19383720
  14. Nonprimed and DYRK1A-primed GSK3 beta-phosphorylation sites on MAP1B regulate microtubule dynamics in growing axons. PMID:19549690
  15. A search of Dyrk1A binding proteins in rat brain finds at least three endocytic accessory proteins that can function as kinase binding proteins. Multiple proteins associated with brain clathrin-coated vesicles are phosphorylated by Dyrk1A. PMID:19722700
