Reconstitution
We recommend that this vial be briefly centrifuged prior to opening to bring the contents to the bottom. Please reconstitute protein in deionized sterile water to a concentration of 0.1-1.0 mg/mL.We recommend to add 5-50% of glycerol (final concentration) and aliquot for long-term storage at -20°C/-80°C. Our default final concentration of glycerol is 50%. Customers could use it as reference.
Storage Condition
Store at -20°C/-80°C upon receipt, aliquoting is necessary for mutiple use. Avoid repeated freeze-thaw cycles.

Description

Asialoglycoprotein receptor 1 mediates the endocytosis of desialylated glycoproteins in hepatocytes, a pathway central to glycoprotein clearance and targeted drug delivery to the liver. This partial construct spanning residues 61–284 encompasses the carbohydrate recognition domain and demonstrates robust conformational integrity, binding an anti-ASGR1 recombinant antibody with a sub-nanomolar affinity constant of 0.12 nM as measured by MetaSPR and an EC50 of 2.336–2.712 ng/mL in functional ELISA. Mammalian expression ensures native post-translational modifications critical for lectin activity, supporting use in ligand-binding assays, competitive inhibition screens for glycan-targeted therapeutics, and therapeutic antibody epitope mapping where authentic receptor conformation is essential. The protein meets quality thresholds commonly required for SPR and BLI affinity characterization, with purity exceeding 95% by SDS-PAGE and endotoxin levels below 1.0 EU/μg.

Mediates the endocytosis of plasma glycoproteins to which the terminal sialic acid residue on their complex carbohydrate moieties has been removed. The receptor recognizes terminal galactose and N-acetylgalactosamine units. After ligand binding to the receptor, the resulting complex is internalized and transported to a sorting organelle, where receptor and ligand are disassociated. The receptor then returns to the cell membrane surface.
