Target Background

Function(From Uniprot)
Plasma membrane-anchored serine protease that participates in proteolytic cascades of relevance for the normal physiologic function of the prostate. Androgen-induced TMPRSS2 activates several substrates that include pro-hepatocyte growth factor/HGF, the protease activated receptor-2/F2RL1 or matriptase/ST14 leading to extracellular matrix disruption. In addition, activates trigeminal neurons and contribute to both spontaneous pain and mechanical allodynia.; (Microbial infection) Essential for spread and pathogenesis of influenza A virus (strains H1N1, H3N2 and H7N9) and is involved in proteolytic cleavage and activation of hemagglutinin (HA) protein which is essential for viral infectivity.
Gene References into Functions
  1. Study shows that Influenza A (IAV) possessing a monobasic cleavage site in the haemagglutinin (HA) replicates poorly in TMPRSS2 knockout mice due to insufficient HA cleavage. On the contrary, IBV successfully underwent HA cleavage in TMPRSS2 knockout mice, and that the mouse-adapted strain was fully pathogenic. These data demonstrate a clear difference between IAV and IBV in their molecular mechanisms for spreading. PMID:27389476
  2. DPP4:CD9:TTSP as the protein complexes are necessary for early efficient MERS-coronavirus entry. PMID:28759649
  3. Genetic inhibition of TMPRSS2-ERG junction oncogene in prostate cancer by means of siRNA has strong antineoplastic effect in a mouse model and in vitro. PMID:25933120
  4. The androgen-regulated protease TMPRSS2 activates a proteolytic cascade involving components of the tumor microenvironment and promotes prostate cancer metastasis PMID:25122198
  5. TMPRSS2 as a host cell factor essential for viral spread and pathogenesis of mono-basic H1N1 and H3N2 influenza A viruses. PMID:24348248
  6. These results demonstrate that TMPRSS2 expression is essential for influenza A virus replication in vivo. PMID:24600012
  7. These data demonstrate that TMPRSS2 is a host factor that is essential for pneumotropism and pathogenicity of H7N9 and H1N1 influenza virus in mice. PMID:24522916
  8. Loss of TMPRSS2 serine protease activity does not influence fertility, reduce survival, result in prostate hyperplasia or carcinoma, or alter prostatic luminal epithelial cell regrowth following castration and androgen replacement. PMID:16428450
