Target Background

Function(From Uniprot)
Mediates, with Gag-Pol polyprotein, the essential events in virion assembly, including binding the plasma membrane, making the protein-protein interactions necessary to create spherical particles, recruiting the viral Env proteins, and packaging the genomic RNA via direct interactions with the RNA packaging sequence (Psi).; Targets the polyprotein to the plasma membrane via a multipartite membrane-binding signal, that includes its myristoylated N-terminus. Matrix protein is part of the pre-integration complex. Implicated in the release from host cell mediated by Vpu. Binds to RNA.; Forms the conical core that encapsulates the genomic RNA-nucleocapsid complex in the virion. Most core are conical, with only 7% tubular. The core is constituted by capsid protein hexamer subunits. The core is disassembled soon after virion entry. Host restriction factors such as TRIM5-alpha or TRIMCyp bind retroviral capsids and cause premature capsid disassembly, leading to blocks in reverse transcription. Capsid restriction by TRIM5 is one of the factors which restricts HIV-1 to the human species. Host PIN1 apparently facilitates the virion uncoating. On the other hand, interactions with PDZD8 or CYPA stabilize the capsid.; Encapsulates and protects viral dimeric unspliced genomic RNA (gRNA). Binds these RNAs through its zinc fingers. Acts as a nucleic acid chaperone which is involved in rearangement of nucleic acid secondary structure during gRNA retrotranscription. Also facilitates template switch leading to recombination. As part of the polyprotein, participates in gRNA dimerization, packaging, tRNA incorporation and virion assembly.; Plays a role in budding of the assembled particle by interacting with the host class E VPS proteins TSG101 and PDCD6IP/AIP1.
Gene References into Functions
  1. The differential requirement for adaptor protein complexes confirmed that HIV-1 and HIV-2 Gag have distinct cellular trafficking pathways, and that HIV-2 particles may be more susceptible to degradation prior to release. PMID:27392064
  2. These data on HIV-2 indicate that the role of the matrix domain in the nucleic acid chaperone activity of Gag differs not only between, but also within, retroviral genera. PMID:26987314
  3. These data show that infectious HIV-2 cannot bear the replacement of the reverse transcriptase Gln294 Pro, as it results in substantially reduced HIV-2 replication and fast reversions to the wild-type Gln294 virus. PMID:25965791
  4. This showed that V111I changed the flexibility of the 110-to-115 loop region, which may affect deoxynucleoside triphosphate (dNTP) binding and polymerase activity. PMID:25355888
  5. HIV-2 genome dimerization is required for the correct processing of Gag. PMID:22419802
  6. The N-terminal and C-terminal domains of the integrase were highly conserved. PMID:21114823
  7. analysis of the secondary active domain of HIV-2 NCp8 PMID:17511966
  8. The results show that all modifications of HIV-2 RT Gln294 lead to an increase of the RNase H activity of the enzyme relative to the wild-type. PMID:18625228
  9. C1-C4 region of HIV2 SU glycoprotein impairs utilization of the CCR5 and CXCR4 for viral entry. PMID:19200444
  10. These results suggest that the splicing of both the 5'UTR intron and the C-box element have key roles in regulation of HIV-2 gag translation in vitro and in vivo. PMID:19559029
