Description
SIRPα is a critical immune checkpoint receptor whose interaction with CD47 delivers a "don't eat me" signal that enables tumor immune evasion, making precise characterization of this axis essential for immuno-oncology drug discovery. This recombinant human SIRPα construct covers the extracellular domain (aa 31–370), encompassing the IgV-like ligand-binding region, and is expressed in mammalian cells to preserve native glycosylation and conformational integrity required for faithful receptor-ligand studies. Validated binding to human CD47 by functional ELISA (EC50 of 58.30–85.04 ng/ml) and by LSPR with a KD of 19.1 nM confirms that this protein supports use in SPR/BLI affinity characterization, competitive blocking antibody screening, therapeutic antibody epitope mapping, and small-molecule inhibitor discovery targeting the SIRPα–CD47 checkpoint. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based immune checkpoint assays and as a reliable positive control in binding experiments.
