EPHA2 mediates contact-dependent cell signaling in tumor progression and angiogenesis, making it a priority target for therapeutic antibody and small-molecule inhibitor development. This mammalian-expressed construct spanning residues 24–534 captures the complete extracellular domain, ensuring native glycosylation and disulfide bonding critical for physiological ligand recognition. Functional ELISA validation demonstrates robust binding to its cognate ligand EFNA1 with an EC50 of 5.859–7.758 ng/mL, confirming that the receptor retains the conformational integrity required for ligand-binding assays, competitive inhibition screens, and therapeutic antibody epitope mapping. The protein also binds an anti-EPHA2 recombinant antibody with an EC50 of 36.24–50.53 ng/mL, supporting its use as a positive control in antibody validation workflows and blocking antibody screening campaigns. Purity exceeding 95% by SDS-PAGE and endotoxin levels below 1.0 EU/μg align with standards expected in surface plasmon resonance and bio-layer interferometry experiments for affinity characterization.
