Target Background

Function(From Uniprot)
Plays a role in protein sorting and trafficking between the trans-Golgi network (TGN) and endosomes. Mediates the ARF-dependent recruitment of clathrin to the TGN and binds ubiquitinated proteins and membrane cargo molecules with a cytosolic acidic cluster-dileucine (DXXLL) motif. Mediates export of the GPCR receptor ADRA2B to the cell surface. Required for targeting PKD1:PKD2 complex from the trans-Golgi network to the cilium membrane. Regulates retrograde transport of proteins such as phosphorylated form of BACE1 from endosomes to the trans-Golgi network.

  1. These findings show that the AD-like phenotype of NPC model cells can be partly reverted by promoting a non-amyloidogenic processing of APP through the upregulation of GGA1 supporting its preventive role against AD PMID:24866237

  11. the trafficking of adiponectin through its secretory pathway is dependent on GGA-coated vesicles PMID:16407204
  12. GGA1 prevented APP beta-cleavage products from becoming substrates for gamma-secretase. Direct binding of GGA1 to BACE was not required for these effects, but the integrity of the GAT (GGA1 and TOM) domain of GGA1 was. PMID:17005855
  13. GGA1 alters the proteolytic processing of beta-amyloid precursor protein (APP) and the secretion of APPs and amyloid-beta, suggesting a role of GGA1 in Alzheimer's disease pathogenesis. PMID:17151287
  14. the interaction between the hinge region and the GAE domain underlies the autoregulation of GGA function in clathrin-mediated trafficking through competing with the accessory proteins and the AP-1 complex PMID:17506864
